Biological Background: PLLP Function and Localization
- Plasmolipin (PLLP), also known as plasma membrane proteolipid (PMLP) or TM4SF11, is a tetraspanin family protein that functions as a major component of the myelin sheath.
- PLLP plays an essential role in myelin membrane biogenesis and myelination, ensuring proper insulation of axons (PubMed:26002055). Related references: PMID:26002055
- In epithelial cells, PLLP is a critical regulator of apical endocytosis, which is required for normal epithelial development (By similarity).
- It mediates the basolateral-to-apical trafficking of ICAM-1 from the subapical compartment to the apical cell membrane in hepatic epithelial cells (PubMed:34999972). Related references: PMID:34999972
- The protein localizes to the cell membrane and is specifically enriched in myelin membranes and apical membranes; it is also detected in the Golgi apparatus.
- PLLP is a transmembrane protein with multiple transmembrane helices, and it undergoes phosphorylation, as indicated by proteomics identification.
- The calculated molecular weight of human PLLP is approximately 20 kDa (UniProt: Q9Y342).
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning the N-terminal region (aa 1-100) of human PLLP. This sequence shows high conservation with mouse and rat orthologs, consistent with the reported cross-reactivity.
- For Western blotting, a band near 20 kDa is expected, corresponding to the predicted molecular weight. Endogenous expression levels may vary among tissues and cell lines.
- In immunofluorescence/ICC, PLLP is anticipated to exhibit membrane-associated staining patterns, particularly in myelin-forming cells and polarized epithelial cells.
- For ELISA applications, the peptide design suggests potential utility for detecting PLLP in cell lysates or tissue extracts; consider using a recombinant protein or peptide as a positive control.
- It is recommended to pre-validate the antibody in the relevant sample system and include appropriate negative controls to confirm specificity.
