Biological Background: PLIN5 Function and Localization

• Perilipin-5 (PLIN5), also known as lipid droplet-associated protein PAT-1, myocardial LD protein, or lipid storage droplet protein 5, is encoded by the Plin5 gene in mouse. It belongs to the perilipin family of lipid droplet coat proteins.
• PLIN5 is a lipid droplet-associated protein that maintains the balance between lipogenesis and lipolysis, and regulates fatty acid oxidation in oxidative tissues. It recruits mitochondria to the surface of lipid droplets and coordinates the storage and release of fatty acids.
• The protein acts as a scaffolding platform for key lipolytic enzymes ABHD5, PNPLA2, and LIPE, reducing PNPLA2 triacylglycerol hydrolase activity by sequestering it on lipid droplets. PKA-mediated phosphorylation promotes lipolysis by releasing ABHD5 and facilitating LIPE interaction.
