Biological Background: Plasminogen Function and Localization
- Plasminogen (PLG) is the zymogen form of plasmin, a serine protease that primarily degrades fibrin, the main component of blood clots (PubMed:6094526, PubMed:6919539). Upon activation, plasmin also cleaves other clot components such as thrombospondin-1 and von Willebrand factor (PubMed:24449821, PubMed:7679575). - Plasminogen contains multiple kringle domains and is activated by cleavage into the heavy and light chains to form active plasmin. Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 20-300) of human plasminogen, covering the PAN domain and kringle structures. This region is well-conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blotting, a band at approximately 91 kDa is expected, corresponding to full-length plasminogen; however, activation fragments or glycosylated forms may appear at different sizes. We recommend optimizing sample preparation and gel percentage.
- For ELISA, use the antibody for capture or detection of recombinant or native plasminogen. Titrate the antibody to determine optimal working concentration.
- Cross-reactivity with mouse and rat has been reported; validate in the relevant sample system for your assay.
