Biological Background: Plasminogen Function and Localization

  • Plasminogen (PLG) is the zymogen form of plasmin, a serine protease that primarily degrades fibrin, the main component of blood clots (PubMed:6094526, PubMed:6919539). Upon activation, plasmin also cleaves other clot components such as thrombospondin-1 and von Willebrand factor (PubMed:24449821, PubMed:7679575). Related references: PMID:6094526 PMID:6919539 PMID:24449821 PMID:7679575
  • Plasminogen contains multiple kringle domains and is activated by cleavage into the heavy and light chains to form active plasmin. It plays a role in extracellular matrix degradation, modulating tissue remodeling and inflammation (PubMed:14699093, PubMed:28849762, PubMed:9171346). Related references: PMID:14699093 PMID:28849762 PMID:9171346
  • Plasminogen exhibits antiangiogenic activity, inhibiting neovascularization, possibly through interaction with ATP synthase subunit ATP5F1A (PubMed:10077593, PubMed:7525077, PubMed:9102221). Related references: PMID:10077593 PMID:7525077 PMID:9102221
  • In vitro, plasminogen can cleave complement zymogens such as C1, C4, and C5 (PubMed:6447255). Related references: PMID:6447255
  • During microbial infection, plasminogen binds to Borrelia burgdorferi OspC, potentially aiding bacterial dissemination, and interacts with dengue virus particles to enhance infection (PubMed:31726374). Related references: PMID:31726374
  • Plasminogen is a secreted glycoprotein synthesized predominantly in the liver and present in plasma and other extracellular fluids.
  • The protein undergoes phosphorylation and disulfide bond formation. It is associated with Keywords: Blood coagulation, Fibrinolysis, Hemostasis, Thrombophilia, Tissue remodeling, Zymogen.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 20-300) of human plasminogen, covering the PAN domain and kringle structures. This region is well-conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
  • For Western blotting, a band at approximately 91 kDa is expected, corresponding to full-length plasminogen; however, activation fragments or glycosylated forms may appear at different sizes. We recommend optimizing sample preparation and gel percentage.
  • For ELISA, use the antibody for capture or detection of recombinant or native plasminogen. Titrate the antibody to determine optimal working concentration.
  • Cross-reactivity with mouse and rat has been reported; validate in the relevant sample system for your assay.
