  • It hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate two second messengers: diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). DAG activates protein kinase C (PKC), while IP3 triggers intracellular Ca2+ release.
  • Beyond its phospholipase activity, PLCD4 acts as a non-receptor guanine nucleotide exchange factor (GEF) for the G protein alpha subunit GNAI3, linking it to G protein signaling.
  • PLCD4 is required for the acrosome reaction in sperm during fertilization, likely by mobilizing Ca2+ in response to zona pellucida stimulation.
  • Overexpression of PLCD4 up-regulates the Erk signaling pathway and promotes cell proliferation, indicating a role in cell growth and liver regeneration.
  • Subcellularly, PLCD4 localizes to the cell membrane, nucleus, cytoplasm, and endoplasmic reticulum, consistent with its signaling roles.
