- PLCD4 encodes 1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase delta-4, also known as phospholipase C-delta-4 (PLC-delta-4), a member of the phospholipase C family.
- It hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate two second messengers: diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). DAG activates protein kinase C (PKC), while IP3 triggers intracellular Ca2+ release.
- Beyond its phospholipase activity, PLCD4 acts as a non-receptor guanine nucleotide exchange factor (GEF) for the G protein alpha subunit GNAI3, linking it to G protein signaling.
- PLCD4 is required for the acrosome reaction in sperm during fertilization, likely by mobilizing Ca2+ in response to zona pellucida stimulation.
- Overexpression of PLCD4 up-regulates the Erk signaling pathway and promotes cell proliferation, indicating a role in cell growth and liver regeneration.
- Subcellularly, PLCD4 localizes to the cell membrane, nucleus, cytoplasm, and endoplasmic reticulum, consistent with its signaling roles.
- It is highly expressed in skeletal muscle, kidney, and moderately in intestinal tissue, and also found in corneal epithelial cells.
