Biological Background: PLCB1 Function and Localization
- PLCB1 encodes 1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase beta-1, also designated PLC-beta-1, PLC-154, or phospholipase C-beta-1.
- The enzyme catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messengers diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3), thereby mediating signal transduction downstream of G protein-coupled receptors. Related references: PMID:9188725
- PLCB1 has been reported to regulate endothelial barrier function.
- The protein localizes to the nuclear membrane and cytoplasm.
- PLCB1 is classified as a key transducer in lipid metabolism, involved in lipid degradation and signaling pathways.
- Alternative splicing yields multiple PLCB1 isoforms, contributing to functional diversity.
- Mutations in PLCB1 are associated with epilepsy, and the protein undergoes post-translational modifications including phosphorylation and palmitoylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 917–1216), which may aid in detecting both full-length and C-terminal variants.
- For WB, the predicted molecular weight is ~139 kDa; consider using an appropriate gel percentage and transfer conditions for high-molecular-weight proteins.
- In IHC-P, perform antigen retrieval and validate in relevant tissue samples.
- For ELISA, consider pairing with a capture antibody if used in a sandwich format.
- Cross-reactivity with mouse and rat allows testing in orthologous models; validate species-specific reactivity in the relevant sample system.
