Biological Background: PLC gamma 2 Function and Localization
- PLCG2 (1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase gamma-2) is also known as phosphoinositide phospholipase C-gamma-2, Phospholipase C-IV (PLC-IV), and Phospholipase C-gamma-2.
- Catalyzes the production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3) from phosphatidylinositol 4,5-bisphosphate, initiating downstream signaling cascades.
Related references: PMID:19394299 and PMID:23000145 - Plays a crucial role in transmembrane signaling by linking activated receptors to intracellular effector pathways.
Related references: PMID:23000145 - Localizes to membrane rafts, specialized microdomains that serve as platforms for signal transduction.
- Contains SH2 and SH3 domains, as well as a calcium-binding region, and is classified as a hydrolase, phosphoprotein, and transducer.
- Variants in PLCG2 are associated with immune disorders; the aliases FCAS3 and APLAID reflect documented disease linkages.
Experimental Guidance and Technical Tips
- The immunogen corresponds to an internal region (aa 701–800) of PLCG2, suggesting recognition of full-length protein and possibly uncharacterized isoforms; validation in the relevant sample system is advised.
- Validated for Western blot and ELISA; these applications may be considered for target detection and quantification. Optimize antibody dilution and incubation conditions for specific experimental setups.
- Due to membrane raft localization, effective protein extraction may require lysis buffers containing detergents (e.g., 1% Triton X-100 or RIPA) to solubilize membrane-associated PLCG2.
- For species cross-reactivity testing, both human and mouse samples are indicated; however, independent verification in other species is recommended before use.
