Biological Background: PLC beta 3 (PLCB3) Function and Localization
- PLC beta 3 (PLCB3), also known as phosphoinositide phospholipase C-beta-3 or phospholipase C-beta-3, is a member of the phospholipase C beta family encoded by the PLCB3 gene.
- Catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messengers diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3), which mobilize calcium and activate protein kinase C. Related references: PMID:20966218, PMID:29122926, PMID:37172014
- Functions as a critical transducer in G protein-coupled receptor (GPCR) signaling, activated by G(q)/G(11) alpha subunits upon receptor stimulation. Related references: PMID:20966218, PMID:37991948
- In neutrophils, PLCB3 participates in N-formyl peptide-activated GPCR pathways, promoting RASGRP4 activation via DAG to support functional responses (By similarity).
- Together with OSBPL2, PLCB3 may regulate keratinocyte proliferation and differentiation, likely through modulation of the ERK signaling pathway and cell cycle progression. Related references: PMID:38701954
- Subcellular localization includes the cytoplasm, membrane, and nucleus, reflecting its role in diverse signaling compartments.
- Post-translational modifications include acetylation and phosphorylation; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the C-terminal region (amino acids 1100-1200) of human PLCB3, ensuring target specificity.
- For Western blot (WB), a band at approximately 139 kDa is expected. Consider using reducing SDS-PAGE and standard transfer protocols; validate in your sample lysate.
- For ELISA, direct or sandwich formats can be tested. Coating conditions and detection antibody concentrations may require optimization.
- Since PLCB3 is expressed in many tissues, include appropriate positive and negative controls to confirm specificity.
