Biological Background: PLA2R1 Function and Localization
- PLA2R1 (Secretory phospholipase A2 receptor), also known as PLA2R, CLEC13C, and PLA2G1R, is a transmembrane receptor belonging to the C-type lectin family (CLEC).
- It functions as a receptor for secretory phospholipase A2 (sPLA2), specifically binding sPLA2-IB/PLA2G1B, leading to MAPK cascade activation, cell proliferation, and lipid mediator production.
- The receptor plays a dual role by also acting as an endocytic receptor that internalizes and clears extracellular sPLA2, thereby protecting cells from potent enzymatic activity.
- In podocytes, PLA2R1 binding of sPLA2-IB/PLA2G1B regulates cell survival and glomerular homeostasis; it is highly expressed in kidney and podocytes. Related references: PMID:25335547
