Biological Background: PLA2R1 Function and Localization

  • Acts as a receptor for secretory phospholipase A2-IB (sPLA2-IB) and snake PA2-like toxins, mediating both positive and negative regulatory effects on sPLA2 functions. Binding triggers MAPK cascade leading to cell proliferation, lipid mediator production, and arachidonic acid release (PubMed:7721806, PubMed:15611272). Related references: PMID:7721806 PMID:15611272
  • Functions as an endocytic receptor that rapidly internalizes sPLA2 ligands, playing a critical role in clearing extracellular sPLA2 to protect against its enzymatic activities (PubMed:15611272). Related references: PMID:15611272
  • A soluble circulating form acts as a negative regulator by blocking the biological functions of sPLA2-IB (PubMed:15611272). Related references: PMID:15611272
  • In podocytes, sPLA2-IB binding regulates podocyte survival and glomerular homeostasis, linking PLA2R1 to kidney health (PubMed:25335547). Related references: PMID:25335547
  • Subcellular localization includes the cell membrane and secretion into the extracellular space; the protein is a heavily glycosylated transmembrane receptor with multiple disulfide bonds.
  • Highly expressed in kidney, but also detected in lung macrophages, pancreas, amnion, choriodecidua, and placenta; isoform 2 is expressed at much lower levels.
  • Belongs to the C-type lectin domain family; alternative splicing produces different isoforms.

Experimental Guidance and Technical Tips

  • For Western blot, use a molecular weight marker up to 250 kDa, and note that glycosylation may cause the band to appear at a slightly higher molecular weight than the predicted 169 kDa.
  • For ELISA, the recombinant protein used as immunogen (aa 810-910) may serve as a suitable coating antigen, but appropriate optimization is recommended.
  • Cross-reactivity with mouse and rat suggests suitability for rodent tissue studies; however, optimal antibody dilutions and conditions should be experimentally validated.
  • The immunogen region lies within the extracellular domain, so the antibody likely recognizes the native receptor on the cell surface.
