Biological Background: PLA2G7 Function and Localization
- PLA2G7 is a lipoprotein-associated, calcium-independent phospholipase A2, also known as platelet-activating factor acetylhydrolase (PAF-AH), LP-PLA2, or LDL-PLA2.
- The enzyme hydrolyzes the sn-2 ester bond of phospholipids, with specificity for short-chain fatty acyl groups and oxidized phospholipids. It inactivates the pro-inflammatory mediator platelet-activating factor (PAF) and prevents accumulation of oxidatively truncated phospholipids in LDL particles, playing a key role in modulating inflammatory and oxidative stress responses.

PMID:2040620, PMID:10504265, PMID:10066756, PMID:11590221, PMID:7700381 - In HDL particles, PLA2G7 can hydrolyze phospholipids with long-chain fatty acyl hydroperoxides, thereby protecting the vascular wall from oxylipin accumulation.
Related references:
PMID:17090529 - PLA2G7 catalyzes the release of F2-isoprostanes from membrane phospholipids, which serve as lipid biomarkers of cellular oxidative damage.
Related references:
PMID:16371369 - The enzyme is secreted into the extracellular space and associates with both low-density lipoprotein (LDL) and high-density lipoprotein (HDL) particles in the blood.
- Primarily expressed in plasma and secreted by macrophages.
- UniProt keywords include Glycoprotein, Secreted, Disease variant, and Asthma, indicating its involvement in inflammatory conditions and genetic susceptibility.
- The PLA2G7 gene encodes a member of the group-VIIA phospholipase A2 family.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 172-441 of human PLA2G7, a region likely to generate antibodies that recognize both native and denatured protein forms.
- For Western blot, a band around 50 kDa is expected under reducing conditions; include appropriate positive controls (e.g., plasma or macrophage lysates).
- ELISA protocols may require optimization of coating antigen and antibody dilution; consider performing a checkerboard titration.
- Cross-reactivity with mouse and rat is indicated; however, validate the antibody in the relevant species and sample type before application.
- Upon receipt, store at -20°C. Aliquot to avoid repeated freeze-thaw cycles and ensure prolonged stability.
