Biological Background: PLA2G6 Function and Localization

  • PLA2G6 encodes an 85/88 kDa calcium-independent phospholipase A2 (also known as iPLA2-beta or PNPLA9) that belongs to the patatin-like phospholipase family and plays key roles in phospholipid remodeling, membrane homeostasis, mitochondrial integrity, and signal transduction.
  • It functions as a phospholipase A1/A2, hydrolyzing fatty acyl chains at the sn-1 and sn-2 positions of glycerophospholipids with preference for sn-2, and also exhibits lysophospholipase activity. Related references: PMID:10092647, PMID:10336645, PMID:20886109
  • Under oxidative stress, PLA2G6 remodels mitochondrial phospholipids by preferentially hydrolyzing oxidized polyunsaturated fatty acyl chains from cardiolipins to repair oxidized lipids, thereby maintaining mitochondrial integrity. Related references: PMID:23533611
  • It possesses thioesterase activity toward fatty-acyl CoA, facilitating fatty acid transport and beta-oxidation in mitochondria, particularly in skeletal muscle. Related references: PMID:20886109
  • PLA2G6 regulates monocyte chemotaxis by controlling F-actin polymerization at pseudopods in response to MCP1/CCL2. Related references: PMID:18208975
  • Alternative splicing produces multiple isoforms with distinct tissue distribution; some isoforms may act as negative regulators due to loss of the catalytic domain.
  • Subcellular localizations include the cytoplasm, cell membrane, mitochondria, and pseudopodia, consistent with its roles in membrane dynamics and signal transduction.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 151–250) lies within a conserved region of PLA2G6; the antibody may recognize multiple isoforms including the catalytically active forms.
  • For western blotting, expect a band near the reported 90 kDa molecular weight, noting that UniProt lists an 85/88 kDa form as well due to potential post‑translational modifications or isoform variation.
  • Cross‑reactivity with rat has been reported; validate the antibody in your specific sample matrix and consider species‑specific sequence alignment if working with other organisms.
  • ELISA is a validated application; consider using this antibody in sandwich or indirect ELISA formats with appropriate secondary detection systems.
