Biological Background: PLA2G4A Function and Localization
- Cytosolic phospholipase A2 (cPLA2α) preferentially hydrolyzes sn-2 arachidonoyl group from membrane phospholipids, providing arachidonic acid for eicosanoid biosynthesis via the cyclooxygenase pathway. Related references: PMID:10358058 PMID:17472963 PMID:18451993
- PLA2G4A also exhibits calcium-independent phospholipase A2 and lysophospholipase activities in the presence of phosphoinositides. Related references: PMID:12672805
- Has O-acyltransferase activity, transferring fatty acyl chains from phospholipids to glycerol, potentially contributing to monoacylglycerol synthesis. Related references: PMID:7794891
- Localizes to the cytoplasm, Golgi apparatus membrane, and nuclear envelope.
- Expressed in various cells and tissues, including macrophages, neutrophils, fibroblasts, lung endothelium, and platelets.
- Plays a role in embryo implantation and parturition through prostanoid production (By similarity).
- Involved in the alternative pathway of eicosanoid biosynthesis by hydrolyzing sn-2 fatty acyl chain of eicosanoid lysophospholipids to release free bioactive eicosanoids. Related references: PMID:27642067
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region (aa 550-749) of human PLA2G4A, which may confer reactivity with both full-length and potential fragments.
- For Western blot, consider using whole cell lysates from human or mouse cells known to express PLA2G4A (e.g., macrophages). The predicted molecular weight is 85 kDa.
- This antibody may be suitable for sandwich ELISA when paired with a detection antibody recognizing a non-overlapping epitope. Validate in the relevant sample system.
