Biological Background: PLA2G2D Function and Localization
- Group IID secretory phospholipase A2 (PLA2G2D; also known as PLA2IID) is a calcium-dependent secreted enzyme that hydrolyzes phospholipids to release free fatty acids and lysophospholipids, exerting anti-inflammatory and immunosuppressive functions. Related references: PMID:10455175 PMID:10681567
- Shows substrate preference for phosphatidylethanolamines and phosphatidylglycerols over phosphatidylcholines. Related references: PMID:10455175
- In lymph nodes, selectively hydrolyzes diacyl and alkenyl forms of phosphatidylethanolamines, releasing omega-3 PUFAs that act as precursors for anti-inflammatory resolvins and drive resolution of inflammation.
- Promotes regulatory T cell (Treg) differentiation and immune tolerance via a catalytic-independent mechanism.
- Has bactericidal activity against Gram-positive bacteria by directly hydrolyzing bacterial membrane phospholipids.
- Secreted protein highly expressed in pancreas and spleen, with moderate levels in colon, thymus, placenta, small intestine, and prostate.

- The immunogen covers amino acids 21-145, representing the mature secreted form of PLA2G2D after signal peptide cleavage. This design may enhance detection of the endogenous secreted protein.
- For Western blot, consider using pancreas or spleen lysates as positive controls based on reported high expression of PLA2G2D.
- In IHC-P, include appropriate antigen retrieval and validate staining patterns in known expression sites such as pancreas or lymphoid tissues.
- For IF/ICC, cells that secrete PLA2G2D or immune cells undergoing activation may serve as relevant models; consider co-staining with established secretion markers.
