Biological Background: PLA2G2D Function and Localization

  • Group IID secretory phospholipase A2 (PLA2G2D; also known as PLA2IID) is a calcium-dependent secreted enzyme that hydrolyzes phospholipids to release free fatty acids and lysophospholipids, exerting anti-inflammatory and immunosuppressive functions.   • Shows substrate preference for phosphatidylethanolamines and phosphatidylglycerols over phosphatidylcholines. Related references: PMID:10455175
  • In lymph nodes, selectively hydrolyzes diacyl and alkenyl forms of phosphatidylethanolamines, releasing omega-3 PUFAs that act as precursors for anti-inflammatory resolvins and drive resolution of inflammation.
  • Promotes regulatory T cell (Treg) differentiation and immune tolerance via a catalytic-independent mechanism.
