Biological Background: PLA2G2A Function and Localization
- PLA2G2A encodes a secreted calcium-dependent phospholipase A2 that hydrolyzes the sn-2 ester bond of phospholipids, with preference for phosphatidylethanolamines and phosphatidylglycerols over phosphatidylcholines, playing a central role in lipid mediator generation and membrane remodeling. Related references: PMID:10455175 PMID:10681567 PMID:2925633
- The protein displays potent bactericidal activity against Gram-positive bacteria by directly hydrolyzing bacterial membrane phospholipids, contributing to innate antimicrobial defense. Related references: PMID:10358193 PMID:11694541
- During sterile inflammation, PLA2G2A targets extracellular mitochondrial phospholipids released from activated platelets, liberating arachidonic acid for eicosanoid synthesis and promoting the release of damage-associated molecular patterns that amplify the immune response. Related references: PMID:25082876
- The enzyme also acts as a ligand for integrins such as ITGAV:ITGB3, ITGA4:ITGB1, and ITGA5:ITGB1, binding to a site distinct from the classical RGD-binding site and inducing conformational changes that enhance cell adhesion and proliferation. Related references: PMID:18635536 PMID:25398877
- PLA2G2A participates in N-acyl ethanolamine biosynthesis by hydrolyzing N-acyl phosphatidylethanolamines to N-acyl lysophosphatidylethanolamines, which are further processed to bioactive lipid amides regulating energy metabolism and inflammation. Related references: PMID:14998370
- Subcellularly, the protein is secreted, associated with the cell membrane, and localized to the mitochondrial outer membrane, reflecting its dual roles in extracellular signaling and organelle membrane dynamics.
- It is expressed in a wide range of tissues including heart, kidney, liver, lung, pancreas, placenta, skeletal muscle, prostate, ovary, colon, and small intestine, with notable expression in platelets, but is absent from lymphoid organs and brain. Related references: PMID:10455175 PMID:10681567 PMID:25082876
- Key structural and functional features include calcium binding, disulfide bonds essential for stability, and multiple functional domains driving antimicrobial, bacteriolytic, and inflammatory activities.
Experimental Guidance and Technical Tips
- For western blot (WB) analysis, consider using denaturing and reducing conditions with a positive control that expresses PLA2G2A; the expected molecular weight is approximately 16 kDa.
- In ELISA applications, ensure optimal coating with full-length native or recombinant PLA2G2A, and validate detection in the relevant sample matrix.
- Cross-reactivity has currently been demonstrated only for human; performance with other species should be experimentally verified.
- As a rabbit polyclonal antibody raised against amino acids 21–144 of human PLA2G2A, it may recognize multiple epitopes; consider lot-specific titration for consistent results.
