Biological Background: PKM2 Function and Localization
- PKM2 catalyzes the final rate-limiting step of glycolysis, converting phosphoenolpyruvate to pyruvate and generating ATP. Its activity is allosterically regulated, and the equilibrium between active tetramer and less active dimer controls metabolic flux. Related references: PMID:15996096, PMID:1854723, PMID:20847263
- The glycolytic enzyme also moonlights as a protein kinase in the nucleus, phosphorylating STAT3 at Tyr-705 and histone H3 at Thr-11 to regulate transcription. Related references: PMID:18191611, PMID:21620138, PMID:22056988, PMID:22306293, PMID:22901803, PMID:24120661
- Nuclear PKM2 promotes transcription of VEGFA alongside NF-kB subunits, enhancing angiogenesis, and drives CD274 expression in specific immune contexts. Related references: PMID:33314660
- PKM2 plays a critical role in tumor cell proliferation and survival; its expression is upregulated in many cancers, making it a key metabolic and regulatory hub in tumorigenesis. Related references: PMID:18337823, PMID:22901803, PMID:26787900
- The protein is predominantly cytoplasmic but translocates to the nucleus upon signaling (e.g., EGFR activation) to exert its non-metabolic functions.
- PKM2 is specifically expressed in proliferating cells, including embryonic stem cells and cancer cells, and can be replaced by the adult PKM1 isoform in differentiated tissues. Related references: PMID:18337823, PMID:20847263
- Post-translational modifications, including acetylation, phosphorylation, and hydroxylation, fine-tune its activity and subcellular localization.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 11-221 of PKM2 (NP_001193728.1), a region spanning the N-terminal domain. Consider testing a range of antibody dilutions (e.g., 1:500–1:5000 for WB) and optimizing blocking conditions for best signal-to-noise ratio.
- For IF/ICC, fix and permeabilize cells appropriately, and validate the staining pattern against known PKM2 subcellular distribution in the relevant cell type.
- When performing ELISA, include proper positive and negative controls to confirm antibody reactivity and specificity.
- Due to cross-reactivity with mouse and rat PKM2, the antibody may be suitable for detecting the target in rodent models; however, confirm by sequence alignment and pilot experiments.
