Biological Background: Polycystin-2 Function and Localization
- Polycystin-2 (PC2, TRPP2, APKD2) is encoded by the PKD2 gene and belongs to the transient receptor potential (TRP) channel family.
- Functions as a nonselective cation channel, operating as a homotetramer or forming heteromers with PKD1; the PKD1/PKD2 complex exhibits higher Ca2+ permeability.
Related references: PMID:11854751, PMID:11991947, PMID:15692563, PMID:31441214, PMID:33164752 - Localizes to multiple subcellular sites, including the primary cilium membrane, endoplasmic reticulum, plasma membrane, and cytoplasmic vesicles, with compartment-specific functions.
- In the primary cilium, acts as a mechanosensor of fluid flow, mediating cation influx (K+, Na+, Ca2+) and playing a role in renal epithelial function and left-right axis specification.
Related references: PMID:27071085 - Regulates intracellular Ca2+ signaling by interacting with TRPV4, TRPC1, IP3R, and RYR2 channels; in cardiomyocytes, modulates Ca2+ release from ryanodine receptors.
Related references: PMID:11854751, PMID:11991947, PMID:27214281 - Highly expressed in fetal and adult kidney (distal tubules, collecting ducts), ovary, testis, and small intestine; also detected in placental syncytiotrophoblasts.
Related references: PMID:10770959, PMID:26269590 - Post-translationally modified by phosphorylation, methylation, and N-glycosylation; contains disulfide bonds and a coiled-coil domain.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 731–830 of human PKD2; ensure that the peptide sequence is conserved across species if using in mouse or rat models.
- For Western blot, a band around 110 kDa is expected. Consider using reducing conditions and loading appropriate positive controls (e.g., human kidney lysate, HEK293 cells overexpressing PKD2).
- For ELISA, the immunizing peptide can serve as a positive coating antigen. Titrate the antibody and optimize detection conditions.
- Cross-reactivity with mouse and rat is reported; however, validation in the specific experimental system is recommended.
