Biological Background: PKC zeta Function and Localization
- Protein identity: Protein kinase C zeta type (nPKC-zeta) is an atypical, calcium- and diacylglycerol-independent serine/threonine kinase.
- Core signaling: Functions downstream of PI3K to activate MAP2K1/MEK1-MAPK1/ERK2 and MAP2K5/MEK5-MAPK7/ERK5 cascades, driving mitogenic signaling and cell proliferation.
- NF-kappa-B pathway: Participates in TNF-dependent NF-kappa-B activation by phosphorylating IKBKB and through complex formation with SQSTM1/p62, RIPK1, and TRAF6, and phosphorylates RELA to relieve SETD6-dependent repression.
- Insulin signaling: Required for insulin-dependent AKT3 activation and contributes to SLC2A4/GLUT4 translocation and glucose transport in adipocytes.
- Cell polarity and differentiation: In migrating astrocytes, complexes with PARD6A and CDC42 to establish cell polarity; stimulates neuronal differentiation via FEZ1 interaction.
- Inflammatory and immune function: Essential for Th2 differentiation, JAK1 activation, and STAT6 phosphorylation; implicated in allergic airway inflammation.
- Subcellular localization: Found in the cytoplasm, endosomes, cell junctions, and membrane.
- Tissue expression: Highly expressed in brain and present at lower levels in lung, kidney, and testis.
- Additional substrate phosphorylation: Phosphorylates VAMP2 in vitro and activates LRRK1, a kinase that phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:17313651, PMID:36040231
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 476-547 in the C-terminal region of human PKC zeta. This antibody may detect full-length protein; consider validating specificity in samples expressing potential splice variants or related PKC isoforms.
- For Western blotting, a band at approximately 68 kDa is expected. Use positive control lysates from brain tissue or cell lines with confirmed PKC zeta expression (e.g., HeLa, HEK293).
- For ELISA applications, the antibody can be used in direct or indirect formats; pairing with a matched detection antibody is recommended for sandwich ELISA.
- As a polyclonal antibody, lot-to-lot consistency should be assessed in critical assays. Pre-adsorption may reduce non-specific binding if necessary.
