Biological Background: PKC zeta Function and Localization

  • Protein kinase C zeta (PKC zeta, PRKCZ) is an atypical protein kinase C (aPKC) isoform that is calcium- and diacylglycerol-independent, belonging to the serine/threonine kinase family. It is encoded by the PRKCZ gene and plays critical roles in signal transduction pathways regulating cell growth, survival, and inflammation.
  • PKC zeta functions downstream of phosphatidylinositol 3-kinase (PI3K) and participates in the mitogen-activated protein (MAP) kinase cascade. It mediates insulin signaling and contributes to glucose transport via SLC2A4/GLUT4 translocation in adipocytes, and activates MAP2K1/MEK1-MAPK1/ERK2 signaling independently of RAF1 following mitogenic stimuli.
  • A key role of PKC zeta is in the activation of the transcription factor NF-kappa-B. Through interaction with SQSTM1/p62 and adapters like RIPK1 and TRAF6, it phosphorylates and activates IKBKB kinase, leading to degradation of NF-kappa-B inhibitors and promoting inflammatory gene expression. It also phosphorylates the RELA subunit at Ser-311, relieving SETD6-mediated repression.
  • PKC zeta is involved in cell polarity establishment. In migrating astrocytes, it forms a complex with PARD6A and is recruited by CDC42 to coordinate microtubule motor and dynein function. This polarity function is crucial for directed cell migration and asymmetric cell division.
  • In neuronal contexts, PKC zeta associates with FEZ1 to stimulate neuronal differentiation in PC12 cells, and is required for the late phase of long-term potentiation (LTP) in hippocampal CA1 cells and long-term memory maintenance.
  • PKC zeta phosphorylates various substrates: it phosphorylates VAMP2 in vitro and also phosphorylates and activates LRRK1, which subsequently phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:17313651 PMID:36040231
  • Subcellularly, PKC zeta is found in the cytoplasm, endosomes, cell junctions, and membrane, consistent with its roles in signaling and trafficking. It is expressed broadly, with highest expression in brain and lower levels in lung, kidney, and testis.
  • The protein undergoes alternative promoter usage and alternative splicing, resulting in isoforms. It contains an ATP-binding domain, a zinc-finger (phorbol ester/DAG-type), and multiple phosphorylation sites. Its molecular weight is approximately 67.7 kDa, but observed bands at 46 kDa, 56 kDa, and 67 kDa likely correspond to isoforms or post-translational modifications.

  • The immunogen is a recombinant fusion protein corresponding to the N-terminal 100 amino acids of human PKC zeta (NP_002735.3). When performing western blot (WB), consider that multiple isoforms or post-translational modifications may result in bands at 46 kDa, 56 kDa, or 67 kDa; validate the observed molecular weight in your sample type.
  • For ELISA applications, ensure that the antibody recognizes the native conformation or linear epitopes as required; perform titration experiments to optimize antibody concentration.
  • Since the antibody is reactive with human PKC zeta, it is suitable for human-derived samples. Cross-reactivity with other species is not reported, so validation is recommended if using non-human systems.
