Biological Background: PKC mu Function and Localization

  • Protein identity and aliases: PKC mu, officially designated serine/threonine-protein kinase D1 (PRKD1), is also known as protein kinase D (PKD), nPKC-mu, and PKCM. It belongs to the protein kinase D family and contains cysteine-rich zinc-finger domains typical of the PKC superfamily.
  • DAG-mediated signaling and kinase activity: PKC mu converts transient diacylglycerol (DAG) signals into sustained physiological responses by phosphorylating EGFR, RIN1, and other substrates, thereby regulating MAPK8/JNK1, Ras, and MAPK1/3 (ERK1/2) signaling cascades. These pathways control cell proliferation, differentiation, and survival. Related references: PMID:10764790 PMID:12505989 PMID:12637538 PMID:10523301
  • Golgi membrane integrity and trafficking: PKC mu acts downstream of heterotrimeric G proteins to maintain Golgi structural integrity and regulates fission of transport vesicles at the trans-Golgi network (TGN), ensuring proper secretory protein trafficking to the plasma membrane.
  • Cell survival and apoptosis regulation: Under oxidative stress, phosphorylation at Tyr-463 by SRC-ABL1 promotes cell survival via IKK/NF-kappa-B activation. Conversely, caspase-3 cleavage at Asp-378 during genotoxic stress enhances its kinase activity and sensitizes cells to apoptosis. Related references: PMID:12505989 PMID:10764790
  • Cell migration and osteoblast differentiation: PKC mu controls cell migration by regulating integrin recycling and focal adhesion turnover. In osteoblasts, it mediates BMP2-induced nuclear export of HDAC7, relieving repression of RUNX2 to promote differentiation. Related references: PMID:18509061
  • Cardiac hypertrophy and angiogenesis: In cardiomyocytes, PKC mu phosphorylates HDAC5, triggering its nuclear export and MEF2-dependent gene expression that drives pathological hypertrophy. During VEGF-induced angiogenesis, it similarly regulates HDAC7 nuclear export to promote endothelial cell migration and tube formation. Related references: PMID:18332134 PMID:19211839
  • Innate immunity and inflammatory responses: PKC mu directly binds and phosphorylates Toll-like receptor 5 (TLR5) upon flagellin stimulation, leading to MAPK14/p38 activation and inflammatory cytokine production. It also phosphorylates NLRP3 to facilitate inflammasome assembly. Related references: PMID:17442957

Experimental Guidance and Technical Tips

  • The immunogen used spans amino acids 200–300 of human PKC mu (Q15139). This region is conserved in mouse and rat (as indicated by cross-reactivity), but sequence verification in your target species is advised for optimal assay design.
  • For Western blotting, expect a band at approximately 102 kDa. High-molecular-weight proteins may require longer transfer times or optimized blotting conditions; loading sufficient protein and using fresh samples can reduce degradation artifacts.
  • In ELISA applications, the antibody’s performance can be influenced by antigen coating concentration and detection system. Titration experiments and inclusion of appropriate controls are recommended.
  • As a rabbit monoclonal antibody, it offers batch-to-batch reproducibility and high affinity, making it suitable for quantitative assays when carefully validated.
  • Cross-reactivity with mouse and rat has been reported; however, the extent of reactivity may vary between applications. Confirm specificity in the relevant tissue or cell context.
