Biological Background: PKC gamma Function and Localization
- Protein kinase C gamma type (PKCγ) is a serine/threonine kinase encoded by the PRKCG gene, belonging to the protein kinase C family. It is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent enzyme.
- Plays pivotal roles in neuronal cells, including regulation of glutamate receptors (GRIA4 and GRIN1), modulation of synaptic plasticity, long-term potentiation (LTP), memory acquisition, and learning.
- Involved in opioid receptor signaling and pain pathways; contributes to morphine tolerance, neuropathic pain, and desensitization of mu-type opioid receptors.
- Mediates ethanol sensitivity and tolerance by modulating GABA(A) receptors, affecting behavioral responses to alcohol.
- Functions in cerebral ischemia: modulates neurotransmission and cell survival, and insulin-induced necrosis inhibition to minimize ischemic injury. Also activated by oxidative stress, leading to connexin-43 phosphorylation and gap junction disassembly.
- Phosphorylates p53/TP53, promoting p53-dependent apoptosis in response to DNA damage.
- Stabilizes the core circadian clock component BMAL1 by interfering with its ubiquitination, contributing to phase resetting of the cerebral cortex clock during restricted feeding.
- Phosphorylates and activates LRRK1, which then phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:36040231
- Subcellular localization: cytoplasm, perinuclear region, cell membrane, synapse, synaptosome, and dendrites. Specifically expressed in Purkinje cells of the cerebellar cortex.
