Biological Background: PKC gamma Function and Localization
- Protein kinase C gamma type (PKC gamma, also known as PKCC, PKCG, SCA14, PKCgamma) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine kinase encoded by the PRKCG gene. Alternative splicing may produce multiple isoforms.
- Plays a central role in neuronal signaling by phosphorylating glutamate receptors such as GRIA4/GLUR4 and GRIN1/NMDAR1, thereby regulating synaptic plasticity, long-term potentiation (LTP), and memory processes.
- Modulates opioid receptor signaling in the spinal cord and limbic forebrain, contributing to morphine-induced reinforcing effects, desensitization of mu-type opioid receptors, and chronic neuropathic pain.
- Exhibits neuroprotective functions: during cerebral ischemia, it modulates neurotransmission and cell survival; in response to oxidative stress, it phosphorylates connexin-43 (GJA1) to disassemble gap junctions, and phosphorylates p53/TP53 to promote apoptosis upon DNA damage.
- Localizes to the cytoplasm, perinuclear region, cell membrane, and neuronal projections including synapses, synaptosomes, and dendrites.
- Highly expressed in Purkinje cells of the cerebellar cortex, and is required for the elimination of multiple climbing fibers during cerebellar development.
- Involved in circadian rhythm regulation by stabilizing the core clock component BMAL1 in the cerebral cortex, thereby suppressing its ubiquitination-mediated degradation (by similarity).
- Phosphorylates and activates LRRK1, which in turn phosphorylates RAB proteins involved in intracellular trafficking (PubMed:36040231). Related references: PMID:36040231
