Biological Background: PKC epsilon Function and Localization

  • Protein kinase C epsilon type (PKC epsilon, also known as nPKC-epsilon) is a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase encoded by the PRKCE gene in humans. It belongs to the protein kinase C family and contains a zinc-finger phorbol-ester/DAG-type domain.
  • PKC epsilon plays key roles in regulating cytoskeletal dynamics, including cell adhesion, motility, migration, and cell cycle progression. It mediates integrin-dependent signaling and phosphorylates substrates such as MARCKS and vimentin to control cell spreading and directional transport.
  • This kinase is involved in neuron growth, synaptic plasticity, and ion channel regulation, contributing to neurite outgrowth and presynaptic facilitation. It also modulates pain signaling by phosphorylating TRPV1.
  • PKC epsilon participates in immune responses by phosphorylating TICAM2/TRAM downstream of TLR4, leading to IRF3 activation and cytokine production. It also contributes to cancer cell invasion through STAT3 phosphorylation.
  • In cardiac cells, PKC epsilon regulates myofilament function and excitation-contraction coupling, and its persistent phosphorylation of troponin I is implicated in dilated cardiomyopathy. It also plays a role in cardiomyocyte hypertrophy.
  • PKC epsilon localizes to the cytoplasm, cytoskeleton, cell membrane, perinuclear region, and nucleus, enabling dynamic spatial regulation of its signaling activities.
