Biological Background: PKC-beta Function and Localization
- Protein kinase C beta (PKC-beta) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that plays pivotal roles in B-cell receptor signaling, oxidative stress-induced apoptosis, and transcriptional regulation. - In B-cell activation, PKC-beta mediates canonical NF-kappa-B signaling by phosphorylating CARD11/CARMA1, promoting recruitment of BCL10-MALT1 and IKK complex activation. It also negatively regulates BTK through phosphorylation at Ser-180. Related references: PMID:11598012
- Under oxidative conditions, PKC-beta phosphorylates p66Shc at Ser-36, triggering mitochondrial reactive oxygen species production and apoptosis.
