Biological Background: PKC alpha Function and Localization
- Protein kinase C alpha (PKCα) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase encoded by the PRKCA gene. It belongs to the PKC family and contains characteristic zinc-finger domains and an ATP-binding site.
- PKCα plays a central role in cell proliferation, differentiation, apoptosis, migration, and adhesion. It promotes cell growth via RAF1-mediated MAPK/ERK signaling and can also induce cell cycle arrest by upregulating CDK inhibitors CDKN1A and CDKN1B.
- Exhibits anti-apoptotic activity in cancer cells; for example, it phosphorylates BCL2 to block apoptosis and suppresses p53/TP53-mediated activation of IGFBP3 in glioma cells.
- Highly expressed in many cancers and functions as a tumor promoter, contributing to malignant phenotypes in gliomas and breast carcinomas.
- In the cardiovascular system, negatively regulates myocardial contractility through phosphorylation of cardiac troponin T (TNNT2/CTNT) and mediates cardiomyocyte hypertrophy via MAPK1/3 (ERK1/2) signaling.
- Involved in angiogenesis, platelet aggregation, and inflammation. It regulates endothelial cell migration and VEGFA signaling, and modulates LPS-induced macrophage responses.
- Subcellular localization includes cytoplasm, cell membrane, mitochondrion membrane, and nucleus. Stimulus-dependent translocation occurs, such as movement to lamellipodia upon wounding or to the nucleus during macrophage differentiation.
