Biological Background: PKC alpha Function and Localization

  • PKC alpha (protein kinase C alpha type) is a calcium-activated, phospholipid- and diacylglycerol-dependent serine/threonine-protein kinase belonging to the PKC family. It regulates diverse cellular processes including proliferation, apoptosis, differentiation, migration, and adhesion.
  • Plays dual roles in cell cycle control: promotes proliferation via RAF1/MAPK/ERK activation and CDKN1A upregulation, and also triggers cell cycle arrest with RB1 hyperphosphorylation and CDK inhibitor induction upon PMA stimulation.
  • Exhibits anti-apoptotic functions in cancer: suppresses TP53-mediated IGFBP3 activation in glioma cells and phosphorylates BCL2 in leukemia cells. Highly expressed in various cancers acting as a tumor promoter.
  • Subcellularly localizes to cytoplasm, cell membrane, mitochondrial membrane, and nucleus. Translocates to lamellipodia after wounding, modulating desmosomal adhesion and cell motility.
  • During chemokine-induced CD4+ T cell migration, phosphorylates DOCK8, leading to CDC42 activation (PubMed:28028151). Related references: PMID:28028151
  • In cardiomyocytes, mediates hypertrophic growth via MAPK1/3 pathway and phosphorylates TNNT2, reducing myocardial contractility. Essential for angiogenesis, endothelial migration, and VEGFA signaling.
