Biological Background: PKC alpha + beta + gamma Function and Localization

  • Protein kinase C beta (PKCβ) is a calcium-activated, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that plays a pivotal role in B-cell receptor (BCR) signal transduction. It mediates NF-kB activation by phosphorylating CARD11/CARMA1 and down-modulates BTK function via direct phosphorylation at Ser-180. Related references: PMID:11598012
  • PKCβ acts as a coactivator of androgen receptor (AR)-dependent transcription by being recruited to AR target genes and specifically phosphorylating histone H3 at Thr-6 (H3T6ph), an epigenetic mark that prevents demethylation of H3K4me. Related references: PMID:20228790
  • In insulin signaling, PKCβ phosphorylates glucose transporter SLC2A1/GLUT1 to promote glucose uptake, and it may function downstream of IRS1 to mediate DNA synthesis via the RAF1-MAPK/ERK pathway. Related references: PMID:25982116
  • PKCβ phosphorylates ATF2, promoting cooperation between ATF2 and JUN and thereby activating transcription. Related references: PMID:19176525
  • It phosphorylates KLHL3 in response to angiotensin II signaling, decreasing the interaction between KLHL3 and WNK4 and thus modulating ion homeostasis. Related references: PMID:25313067
  • PKCβ phosphorylates and activates LRRK1, which in turn phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:36040231
  • PKCβ is an activator of ferroptosis: it phosphorylates ACSL4 to promote lipid peroxidation and phosphorylates FOXK1 to stimulate LGALS13 secretion, propagating ferroptosis signaling. Related references: PMID:35027735 PMID:40246981
  • Subcellular localization includes cytoplasm, nucleus, and membrane. The antibody’s immunogen is derived from a conserved region shared by PKC alpha, beta, and gamma, enabling pan-isoform recognition across human, mouse, and rat.

Experimental Guidance and Technical Tips

  • The immunogen peptide corresponds to a conserved sequence (aa 300–500) in PKC alpha, beta, and gamma, which may allow detection of multiple isoforms. Confirm specificity by using isoform-specific controls or knockdown/inhibitor studies if needed.
  • For WB, consider a starting dilution range and optimize based on target abundance in your sample type; the predicted band size is approximately 78 kDa.
  • In ELISA, the antibody can be used for capture or detection; pairing with an appropriate secondary antibody and validating in the relevant species matrix is recommended.
  • Cross-reactivity has been observed with human, mouse, and rat samples; always include appropriate positive and negative controls when analyzing samples from other species.
