Biological Background: PKA C-beta (PRKACB) Function and Localization

  • Encodes the catalytic subunit beta of cAMP-dependent protein kinase (PKA), a serine/threonine kinase central to cAMP-mediated signaling downstream of G protein-coupled receptors (GPCRs). PKA holoenzyme is a tetramer of two regulatory and two catalytic subunits, with the catalytic subunit released upon cAMP binding to initiate phosphorylation of target proteins. Related references: PMID:12420224 PMID:21423175 PMID:31112131
  • Regulates a broad spectrum of cellular processes including proliferation, cell cycle progression, differentiation, microtubule dynamics, chromatin remodeling, nuclear envelope dynamics, intracellular transport, and ion flux. Related references: PMID:12420224 PMID:21423175
  • Functions as a negative regulator of mTORC1 by directly phosphorylating the RPTOR subunit, linking nutrient sensing and growth control. Related references: PMID:31112131
  • Phosphorylates the RNA-binding protein GPKOW, modulating its RNA-binding activity and potentially influencing pre-mRNA processing. Related references: PMID:21880142
  • Exhibits multiple isoforms with distinct tissue distributions: isoform 1 is brain-abundant, isoform 2 predominates in thymus, spleen, and kidney, while isoforms 3 and 4 are brain-specific.
  • Localizes to the cytoplasm, cell membrane, and nucleus, consistent with its roles in signal transduction and transcriptional regulation.
  • Contains N-myristoylation and other post-translational modifications that contribute to membrane targeting and dynamic subcellular redistribution.

Experimental Guidance and Technical Tips

  • Immunogen derived from the C-terminal region (aa 251–351) common to all isoforms; consider verifying isoform detection specificity in your model system.
  • For western blotting, a band around 41 kDa is expected; validate with appropriate positive and negative controls due to potential isoform-specific expression patterns.
  • IHC‑P and IF/ICC may require optimized antigen retrieval and blocking; test signal specificity in tissue/cell types with known PKA C‑beta expression (e.g., brain for isoform 1).
  • ELISA application can be considered for quantitative detection of the protein or antibody titering; include recombinant protein or peptide standards.
  • Cross-reacts with human, mouse, and rat; verify sequence homology for other species if needed.
