Biological Background: PKA C-alpha Function and Localization

  • PKA C-alpha, encoded by PRKACA, is the catalytic subunit alpha of cAMP-dependent protein kinase. Two isoforms exist: isoform 1 is ubiquitously expressed, while isoform 2 is sperm-specific and enriched in pachytene spermatocytes.
  • As a serine/threonine kinase, it phosphorylates numerous substrates in the cytoplasm and nucleus, including NFKB1 (PubMed:15642694), VASP (PubMed:15642694, PubMed:20356841), RYR2 (PubMed:17693412), and ABL1 (PubMed:15178447). These events regulate platelet aggregation, calcium signaling, and sperm capacitation. Related references: PMID:15642694, PMID:20356841, PMID:17693412, PMID:15178447
  • It localizes to multiple compartments, including cytoplasm, nucleus, cell membrane, mitochondria, cilia, and secretory vesicles, reflecting its diverse regulatory roles.
  • In the hedgehog signaling pathway, PKA C-alpha acts as a key inhibitor by phosphorylating GLI transcription factors (PubMed:16705181). This phosphorylation promotes GLI processing and prevents target gene activation, a process antagonized by SMO-mediated sequestration (PubMed:36202993, PubMed:39138140). Related references: PMID:16705181, PMID:36202993, PMID:39138140
  • The protein is myristoylated and contains nucleotide-binding and kinase domains, classifying it as a lipoprotein and transferase. Disease-associated variants are linked to Cushing syndrome.
  • It also phosphorylates CDC25B to prevent meiosis resumption in prophase-arrested oocytes (By similarity) and regulates tight junctions via CLDN3 phosphorylation (PubMed:15905176). Related references: PMID:15905176

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 1-351 of human PKA C-alpha) spans nearly the entire protein, suggesting recognition of both isoforms. Consider validating isoform specificity using isoform-specific samples (e.g., sperm cells for isoform 2).
  • For Western blotting, the expected molecular weight is approximately 40–41 kDa. Use appropriate positive controls (e.g., HeLa, Jurkat lysates) and validate in your target tissue.
  • In immunofluorescence/ICC, nuclear and cytoplasmic staining is expected; verify subcellular localization under your experimental conditions.
  • For ELISA, pair with a suitable capture antibody and confirm performance with recombinant PKA C-alpha or relevant biological samples.
