Biological Background: PKA C-alpha Function and Localization
- PKA C-alpha, encoded by PRKACA, is the catalytic subunit alpha of cAMP-dependent protein kinase. Two isoforms exist: isoform 1 is ubiquitously expressed, while isoform 2 is sperm-specific and enriched in pachytene spermatocytes.
- As a serine/threonine kinase, it phosphorylates numerous substrates in the cytoplasm and nucleus, including NFKB1 (PubMed:15642694), VASP (PubMed:15642694, PubMed:20356841), RYR2 (PubMed:17693412), and ABL1 (PubMed:15178447). - It localizes to multiple compartments, including cytoplasm, nucleus, cell membrane, mitochondria, cilia, and secretory vesicles, reflecting its diverse regulatory roles.
- In the hedgehog signaling pathway, PKA C-alpha acts as a key inhibitor by phosphorylating GLI transcription factors (PubMed:16705181).
