Biological Background: PIWIL2 Function and Localization
- PIWIL2 (Piwi-like protein 2) is an endoribonuclease that plays a central role in spermatogenesis by repressing transposable elements via the piRNA pathway (By similarity).
- Acts as a 'slicer-competent' piRNA endoribonuclease in the ping-pong amplification loop, cleaving primary piRNAs to produce mature piRNAs and facilitating PIWIL4 nuclear localization and secondary piRNA loading (By similarity).
- Essential for meiotic differentiation of spermatocytes and self-renewal of spermatogonial stem cells, indirectly modulating expression of genes like PDGFRB, SLC2A1, and STRA8 (By similarity).
- Overexpression of PIWIL2 inhibits apoptosis and promotes proliferation in tumors, acting as an oncogene. Related references: PMID:16377660
- Represses circadian rhythms by stabilizing core clock components BMAL1 and CLOCK, inhibiting GSK3B-mediated phosphorylation and degradation. Related references: PMID:28903391
- Subcellularly localized in the cytoplasm (UniProt Q8TC59).
- Expressed in adult testis and in most tumors, highlighting its role in germline integrity and cancer.
- Belongs to the Piwi family of Argonaute proteins, characterized by RNA-binding and metal-binding properties.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 100-200 of human PIWIL2 (Q8TC59); for Western blot, consider using testis or tumor cell lysates as positive controls given the tissue-specific expression.
- Cross-reactivity with mouse is confirmed; this antibody may be suitable for mouse spermatogenesis and piRNA pathway studies.
- For ELISA, validate coating and detection conditions empirically, and confirm specificity with appropriate controls.
- Ensure proper handling: shipped on ice bags to maintain stability upon receipt.
