Biological Background: PIWIL1 Function and Localization

  • PIWIL1 (Piwi-like protein 1) is an endoribonuclease that plays a central role in postnatal germ cells by repressing transposable elements via the piRNA pathway, essential for germline integrity.
  • Acts as an endoribonuclease that cleaves transposon messenger RNAs; strongly prefers a uridine in the first position of its guide piRNAs (1U-bias).
  • May function as a negative developmental regulator. Related references: PMID:12037681 PMID:16287078
  • Localizes to the cytoplasm and is required for the formation of chromatoid bodies.
  • Expressed specifically in spermatocytes and spermatids in the testis; also detected in prostate cancer and overexpressed in gastric cancer cells. Isoform 3 is ubiquitously expressed, including in CD34+ hematopoietic progenitor cells.
  • Binds methylated piRNAs and is not involved in the piRNA ping-pong amplification cycle.
  • Required for the stability of some miRNAs and sequesters RNF8 in the cytoplasm until late spermatogenesis; degraded by ubiquitination.
