Biological Background: PITPNM1 Function and Localization
- Membrane-associated phosphatidylinositol transfer protein 1 (PITPNM1, also known as NIR-2 or Drosophila retinal degeneration B homolog) shuttles phosphatidylinositol (PI) between the endoplasmic reticulum and the plasma membrane, maintaining phosphoinositide signaling competence. It mediates PI/phosphatidic acid counter-transport at ER-PM contact sites, essential for sustained receptor-coupled PLC signaling and replenishment of plasma membrane phosphoinositides. - Regulates RHOA activity and plays a role in cytoskeleton remodeling. Related references: PMID:11909959
- Necessary for normal completion of cytokinesis. Related references: PMID:15125835
- Maintains normal diacylglycerol levels in the Golgi apparatus and is required for the structural integrity of the endoplasmic reticulum and Golgi. It is also necessary for protein export from these organelles. Related references: PMID:15723057 PMID:15545272 PMID:11909959
- Subcellular localization includes the cell membrane, Golgi stack membrane, endoplasmic reticulum membrane, lipid droplets, cleavage furrow, and midbody.
- Ubiquitously expressed in human tissues.
- Annotated with keywords highlighting its involvement in calcium and metal binding, protein transport, and its characterization as a phosphoprotein subject to alternative splicing and methylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 110–190 of human PITPNM1. This region lies within the N-terminal portion of the protein and is well conserved; polyclonal antibodies against it may detect multiple splice isoforms.
- For Western blotting, a band near the predicted molecular weight of 135 kDa is expected. Due to potential post-translational modifications or isoform variation, validate with appropriate positive and negative controls in your sample system.
- ELISA procedures may require optimization of coating antigen (recombinant protein or peptide) and antibody titration. Cross-reactivity with mouse and rat orthologs enables multi-species studies.
- As a polyclonal antibody, consider lot-to-lot consistency testing for critical quantitative applications.
