Biological Background: GOPC Function and Localization
- GOPC (Golgi-associated PDZ and coiled-coil motif-containing protein, also known as PIST, CAL, FIG) is a multidomain adaptor protein involved in intracellular protein trafficking and degradation. Related references: PMID:11707463, PMID:14570915, PMID:15358775
- It regulates the cell surface expression and activity of CFTR chloride channels and ASIC3 acid-sensing channels, influencing ion transport.
- GOPC directly interacts with and modulates trafficking of the ADR1B receptor. Related references: PMID:15358775
- Together with the E3 ubiquitin ligase MARCHF2, GOPC mediates ubiquitination and lysosomal degradation of CFTR, leading to intracellular retention. Related references: PMID:23818989
- GOPC has been implicated in the regulation of autophagy.
- Subcellularly, GOPC localizes to the cytoplasm, Golgi apparatus (including trans-Golgi network), postsynaptic density, and dendrites.
- The protein undergoes acetylation and alternative splicing, and its gene is involved in chromosomal rearrangements. GOPC is ubiquitously expressed in human tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 195-454) of human GOPC, encompassing PDZ and coiled-coil domains; this region is conserved across human, mouse, and rat, supporting the reported cross-reactivity.
- For WB, a specific band around 51kDa (predicted molecular weight 50.5kDa) is expected. Consider using positive lysates from human, mouse, or rat tissues/cell lines and optimizing antibody dilution and detection conditions.
- In ELISA, the antibody can detect recombinant or endogenous GOPC; validate performance with appropriate standards and detection reagents.
- Given GOPC's role in CFTR regulation, autophagy, and receptor trafficking, this antibody may be useful in mechanistic studies; for applications beyond WB and ELISA (e.g., IP or IF), independent validation is recommended.
