Biological Background: PIST/GOPC Function and Localization

  • PIST/GOPC (Golgi-associated PDZ and coiled-coil motif-containing protein) is a ubiquitously expressed adaptor protein encoded by the GOPC gene. Alternate names include CAL (CFTR-associated ligand), FIG (fused in glioblastoma), and PIST (PDZ protein interacting specifically with TC10).
  • Functions in intracellular protein trafficking and degradation, influencing the cell surface expression and stability of various transmembrane proteins. Related references: PMID:11707463 PMID:14570915 PMID:15358775
  • Regulates CFTR chloride currents and acid-sensing ion channel ASIC3 currents by modulating their cell surface expression (By similarity).
  • Regulates the intracellular trafficking of the ADR1B receptor. Related references: PMID:15358775
  • Together with the E3 ubiquitin ligase MARCHF2, mediates ubiquitination and lysosomal degradation of CFTR. Related references: PMID:23818989
  • Overexpression leads to CFTR intracellular retention and lysosomal degradation. Related references: PMID:11707463 PMID:14570915
  • Predominantly localizes to the Golgi apparatus membrane and trans-Golgi network, but is also found in the cytoplasm, postsynaptic density, and dendritic projections.
  • May play a role in autophagy (By similarity).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal 1-100 amino acids of human PIST/GOPC. This region is conserved across human, mouse, and rat, supporting the reported cross-reactivity.
  • For Western blotting, a band at approximately 51 kDa is expected. Validate specificity using positive and negative controls expressing or lacking the target.
  • In immunohistochemistry (IHC-P), antigen retrieval conditions may require optimization. Given ubiquitous expression, many tissues are suitable as positive controls; nonetheless, validate in your specific sample system.
  • For immunofluorescence (IF/ICC), permeabilization and fixation conditions should be optimized for Golgi and cytoplasmic detection.
  • ELISA applications may benefit from pairing with a suitable detection antibody; consider checkerboard titrations to determine optimal working concentrations.
  • Always include appropriate controls (e.g., siRNA knockdown or knockout cell lines) to confirm antibody specificity.
