Biological Background: PINK1 Function and Localization
- PINK1 is a serine/threonine-protein kinase that acts as a sensor of mitochondrial damage, phosphorylating proteins to coordinate mitochondrial quality control mechanisms that remove and replace dysfunctional components. - Upon irreversible mitochondrial stress, PINK1 accumulates at the outer mitochondrial membrane, phosphorylates polyubiquitin chains, recruits and activates PRKN, which ubiquitinates OMM proteins to initiate mitophagy. Related references: PMID:14607334, PMID:15087508, PMID:18443288, PMID:19966284

Experimental Guidance and Technical Tips
- Given the immunogen corresponding to amino acids 282–581, the antibody may recognize full-length PINK1 (~63 kDa) and potentially processed forms. For western blotting, validate using appropriate positive controls such as heart or skeletal muscle lysates and consider using mitochondrial enrichment.
- For immunofluorescence (IF/ICC), expect a mitochondrial staining pattern co-localizing with mitochondrial markers like Tom20; include mitophagy inducer (e.g., CCCP) treatments to observe accumulation.
- For IHC-P, test on known high-expression tissues (heart, skeletal muscle, testis) and perform antigen retrieval optimization. Cross-reactivity with mouse and rat allows for model organism studies.
- ELISA applications may be suitable for detecting PINK1 in cell or tissue lysates; however, pairing with validated standards is recommended.
