Biological Background: PIN4 Function and Localization

  • PIN4 encodes two isoforms, isoform 1 (Par14) and isoform 2 (Par17), produced by alternative promoter usage. Both belong to the parvulin family of peptidyl-prolyl cis-trans isomerases (PPIases) and function as rotamases.
  • Isoform 1 (Par14) plays a role in ribosomal RNA processing and ribosome biogenesis. It also shows affinity for tightly bent AT-rich stretches of double-stranded DNA.
  • Isoform 2 (Par17) is a DNA-binding protein that interacts with double-stranded DNA.
  • The protein localizes to multiple subcellular compartments, including the nucleus, nucleolus, cytoplasm, cytoskeleton (spindle), and mitochondria (mitochondrial matrix).
  • Post-translational modifications, such as phosphorylation and ubiquitin-like conjugation, have been identified, suggesting regulation of PIN4 activity.
  • Expression is ubiquitous, with isoform 1 transcripts more abundant than isoform 2. Isoform 2 protein is more stable. Detected in tissues including kidney, liver, blood vessel, brain, mammary gland, skeletal muscle, small intestine, and submandibularis.

Experimental Guidance and Technical Tips

  • For Western blotting, consider optimizing the antibody dilution as a polyclonal reagent. Validate the detection of both Par14 (~14 kDa) and Par17 (~17 kDa) isoforms, as the immunogen (aa 26-156) covers regions present in both.
  • When using ELISA, a recombinant PIN4 protein containing the immunogen sequence may serve as an appropriate standard control. Confirm linearity and sensitivity in the relevant sample matrix.
  • Cross-reactivity with mouse and rat is indicated; however, verify specificity and signal intensity in these species through pilot experiments. Endogenous PIN4 levels and isoform distribution may vary across tissues and cell lines.
