Biological Background: PIM3 Function and Localization
- PIM3 is a serine/threonine-protein kinase and proto-oncogene belonging to the PIM kinase family. It functions as an ATP-binding kinase that phosphorylates target proteins to regulate cell survival and proliferation.
- The kinase prevents apoptosis and promotes protein translation, contributing to tumorigenesis through phosphorylation of BAD, which releases the anti-apoptotic protein Bcl-X(L), and by modulating MYC transcriptional activity.
- PIM3 also negatively regulates insulin secretion by inhibiting MAPK1/3 (ERK1/2) activation via SOCS6, and is involved in energy metabolism through regulation of AMPK activity, MYC, and PPARGC1A protein levels.
- Subcellular localization is primarily cytoplasmic, consistent with its role in signal transduction pathways.
- The protein is classified as a phosphoprotein and is subject to ubiquitin-like conjugation (Ubl), as indicated by proteomics identification and keyword annotations.
- Tissue-specific expression is detected in heart, brain, lung, kidney, spleen, placenta, skeletal muscle, and peripheral blood leukocytes, while expression is low or absent in normal adult endoderm-derived organs (colon, thymus, liver, small intestine) but becomes augmented in premalignant and malignant lesions.
Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight is approximately 36 kDa. Optimize blocking and antibody dilution to minimize background, as polyclonal antibodies may recognize multiple epitopes.
- In ELISA, consider testing the antibody in both direct and capture formats, depending on assay design and required sensitivity.
- Since the immunogen corresponds to the C-terminal region (amino acids 247–326), the antibody is expected to detect full-length PIM3. Validate specificity in human samples, as cross-reactivity with other species has not been characterized.
- As a polyclonal antibody, it may provide robust signal across applications, but each new assay should be validated with appropriate positive and negative controls.
