Biological Background: PIM1 Function and Localization
- PIM1 is a serine/threonine-protein kinase that acts as a proto-oncogene, promoting cell survival and proliferation and providing a selective advantage in tumorigenesis. Related references: PMID:15528381, PMID:1825810, PMID:31548394
- It exerts oncogenic activity by regulating MYC transcriptional activity and phosphorylating/inhibiting proapoptotic proteins BAD, MAP3K5, and FOXO3. Related references: PMID:18593906
- PIM1 phosphorylates MAP3K5, reducing its kinase activity and inhibiting downstream JNK/p38MAPK-mediated caspase-3 activation and apoptosis. Related references: PMID:19749799
- The kinase promotes cell cycle progression at G1-S and G2-M transitions by phosphorylating CDC25A and CDC25C, and regulates CDKN1A and CDKN1B stability and localization. Related references: PMID:16356754, PMID:12431783, PMID:18593906
- PIM1 may influence chromatin structure through phosphorylation of HP1 gamma/CBX3 and positively regulates mTORC1 signaling by phosphorylating and inhibiting DEPDC5. Related references: PMID:10664448, PMID:31548394
- It phosphorylates and activates ABCG2, contributing to drug resistance, and acts as a negative regulator of innate immunity by phosphorylating GBP1. Related references: PMID:18056989, PMID:37797010
- PIM1 localizes to the cytoplasm, nucleus, and cell membrane. It is predominantly expressed in hematopoietic and germline tissues, with isoforms detected in prostate cancer.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human PIM1. This region is conserved across human, mouse, and rat, contributing to cross-reactivity. Verify sequence homologies for other species if needed.
- For Western blotting (WB), a specific band at approximately 36 kDa is expected. Include appropriate positive (e.g., HeLa, Jurkat) and negative controls to confirm specificity.
- For immunohistochemistry (IHC-P), standard antigen retrieval methods may be required. Consider testing multiple retrieval buffers and optimize antibody dilution based on tissue type.
- In ELISA, use recombinant or purified PIM1 protein as the coating antigen. Perform checkerboard titrations to determine optimal antibody and antigen concentrations.
- Due to the protein’s involvement in multiple signaling pathways, results should be validated using complementary approaches such as siRNA-mediated knockdown or overexpression assays.
