  • PIK3CG encodes the catalytic subunit p110γ of class I phosphoinositide 3-kinase (PI3K) gamma, also known as PI3Kgamma or p120-PI3K, and belongs to the PI3K family of lipid kinases that phosphorylate phosphoinositides.
  • The enzyme specifically phosphorylates phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) to generate phosphatidylinositol 3,4,5-trisphosphate (PIP3), a critical second messenger that recruits PH domain-containing proteins like AKT1 and PDPK1 to the plasma membrane to activate downstream signaling cascades.
  • PIK3CG links G protein-coupled receptor (GPCR) activation to PIP3 production and plays essential roles in immune, inflammatory, and allergic responses by modulating leukocyte chemotaxis, polarization, and migration.

  • Tissue expression is highest in the pancreas, skeletal muscle, liver, and heart; genetic variants are associated with immunodeficiency (IMD97).
