Biological Background: PIGU Function and Localization
- PIGU (Phosphatidylinositol glycan anchor biosynthesis class U protein) is a component of the GPI-anchor transamidase (GPI-T) complex that catalyzes the linkage between proproteins and the GPI anchor, essential for GPI-anchored protein biosynthesis. Related references: PMID:12802054, PMID:31353022, PMID:34576938
- PIGU binds the lipid portion of the GPI anchor, facilitating proper complex assembly at the membrane. Related references: PMID:37684232
- It may act as an organizer in the transmembrane layer, recruiting other subunits and being critical for GPI-T complex integrity. Related references: PMID:35165458, PMID:35551457
- Subcellular localization is at the endoplasmic reticulum membrane.
- Alternative splicing yields multiple isoforms; disease variants are associated with epilepsy and intellectual disability.
- Keywords include GPI-anchor biosynthesis, membrane protein, alternative splicing, and disease variant.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1–100) of human PIGU. For IF/ICC, consider permeabilization protocols to access this intracellular epitope.
- In WB, a band near 50 kDa is expected. Validate detection with appropriate positive control lysates from human, mouse, or rat samples.
- For IHC-P, optimize antigen retrieval conditions and antibody titration for your tissue type.
- For ELISA, the antibody may serve as a capture or detection reagent; pair with a validated anti-PIGU antibody conjugate.
- Cross-reactivity with mouse and rat has been reported; verify reactivity if using other species.
