Biological Background: PIGR Function and Localization

  • Polymeric immunoglobulin receptor (PIGR), also known as hepatocellular carcinoma-associated protein TB6, is a transmembrane glycoprotein central to mucosal immune defense.
  • It mediates selective transcytosis of polymeric IgA (pIgA) and IgM across mucosal epithelial cells, binding them at the basolateral surface and transporting the complex to the apical surface for secretion.
  • During transcytosis, the extracellular ligand-binding domain (secretory component) is proteolytically cleaved and released with the immunoglobulin, forming secretory IgA (sIgA) that is anchored to the mucus lining via N-linked glycans. Related references: PMID:12150896
  • In its free form (secretory component), PIGR can act as a non-specific microbial scavenger, directly binding to bacteria and preventing their adhesion to epithelial cells. Related references: PMID:16543244

  • For Western blot, the predicted full-length molecular weight is ~83 kDa; cleavage products (~80 kDa) may also appear. Consider testing reducing and non-reducing conditions due to the presence of disulfide bonds.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), expect staining at the apical surface of mucosal epithelial cells and in secreted material; include appropriate tissue controls.
  • Cross-reactivity with mouse and rat has been confirmed; for other species, verify sequence homology before use.
