Biological Background: PIGQ Function and Localization
- PIGQ (Phosphatidylinositol N-acetylglucosaminyltransferase subunit Q), also known as GPI1 or PIG-Q, is a subunit of the GPI-GnT complex.
- The GPI-GnT complex catalyzes the transfer of N-acetylglucosamine from UDP-N-acetylglucosamine to phosphatidylinositol, the first step in GPI anchor biosynthesis.
- GPI anchors are glycolipids that attach many proteins to the cell surface, essential for normal cellular function.
- Mutations in PIGQ are associated with diseases such as developmental and epileptic encephalopathy 77 (DEE77) and multiple congenital anomalies-hypotonia-seizures syndrome 4 (MCAHS4).
- PIGQ localizes to the membrane and contains transmembrane helices, consistent with its role in GPI synthesis at the endoplasmic reticulum.
- Alternative splicing generates multiple isoforms; the canonical isoform has a molecular weight of approximately 84 kDa.
- Keywords: Alternative splicing, Disease variant, Epilepsy, GPI-anchor biosynthesis, Membrane, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-200) of human PIGQ, which may be conserved across species; cross-reactivity with mouse is observed.
- For Western blotting, consider a starting dilution range based on endogenous expression levels in relevant cell lines; the predicted molecular weight is ~84 kDa.
- The antibody may also be suitable for ELISA; optimize coating and detection conditions in the target sample matrix.
- As PIGQ is a multi-pass membrane protein, consider including mild detergents in lysis buffers to maintain solubility.
- Validate specificity using knockout or knockdown controls if available.
