Biological Background: PIDD Function and Localization
- PIDD (p53-induced death domain-containing protein 1) is also known as leucine-rich repeat and death domain-containing protein (LRDD). It plays a dual role in cell fate decisions downstream of p53, mediating either survival or apoptosis in response to DNA damage and stress. Related references: PMID:10973264 PMID:15073321 PMID:16360037
- It forms the PIDDosome complex with CRADD and CASP2, leading to CASP2 activation and the induction of apoptosis. Related references: PMID:15073321 PMID:17159900
- PIDD interacts with IKBKG and RIPK1 to promote sumoylation and ubiquitination of IKBKG, which is critical for NF-kappa-B transcription factor activation. Related references: PMID:16360037 PMID:17159900
- The protein localizes to both the cytoplasm and nucleus.
- PIDD is ubiquitously expressed across tissues.
- It undergoes acetylation and phosphorylation; contains leucine-rich repeats and a death domain; alternative splicing generates multiple isoforms; mutations are associated with intellectual disability.
- The full-length protein has a calculated molecular weight of approximately 100 kDa (99.7 kDa).
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1-85), conserved across isoforms, so the antibody may detect multiple PIDD variants; validate isoform specificity in your model system.
- For Western blot, a band near 100 kDa is expected; use appropriate positive and negative controls to confirm target identity.
- The antibody is applicable in IHC-P and IF/ICC; optimize fixation, permeabilization, and antigen retrieval steps for your particular samples.
- With reported cross-reactivity to human, mouse, and rat, the antibody can be considered for cross-species studies; however, verify reactivity in your species and tissue context.
- For ELISA, consider pairing with a suitable detection reagent, and validate sensitivity and specificity in your assay format.
