Biological Background: PIAS1/PIAS2 Function and Localization

  • PIAS1 acts as an E3 SUMO ligase and SUMO-tethering factor, catalyzing the SUMOylation of diverse substrates. It functions as a critical transcriptional coregulator that modulates the STAT, p53, and steroid hormone signaling pathways, either activating or repressing gene expression depending on the cellular context.   • PIAS1 binds to A/T-rich DNA sequences in vitro and directly interacts with gene regulatory regions, such as the TAAT box of the MYOD1 promoter, to regulate transcription factor binding independently of its SUMOylation activity. Related references: PMID:15133049
  • PIAS1 localizes primarily to nuclear speckles and PML bodies, with additional presence in the cytoplasm and association with the cytoskeleton, facilitating its roles in both nuclear processes and cytoplasmic signaling.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 552–651 of human PIAS1; when performing western blot, a major band at approximately 76 kDa is expected for both PIAS1 and PIAS2.
  • For WB and IHC-P, consider using positive control lysates from testis or cell lines expressing high levels of PIAS1/2, and validate the antibody with recombinant proteins or siRNA/shRNA knockdown.
  • Optimal antibody dilution should be determined empirically; start with 1:500–1:2000 for WB and 1:100–1:500 for IHC-P under standard conditions.
  • Due to the high conservation of the epitope region, this antibody also detects PIAS2, but it is advisable to confirm specificity in knockout or knockdown models.
  • For ELISA applications, coating antigen may require optimization of peptide or protein capture; consider using the immunogen peptide as a positive control.
