Biological Background: PI4K2B Function and Localization
- PI4K2B, also known as phosphatidylinositol 4-kinase type II-beta, is a lipid kinase that catalyzes the phosphorylation of phosphatidylinositol (PI) to phosphatidylinositol 4-phosphate (PI4P). Related references: PMID:11923287, PMID:12324459
- This reaction is the first committed step in the synthesis of phosphatidylinositol 4,5-bisphosphate (PIP2), a precursor of the second messengers inositol 1,4,5-trisphosphate (InsP3) and diacylglycerol. Related references: PMID:11923287, PMID:12324459
- Together with PI4K2A and type III PI4Ks, PI4K2B contributes to cellular PI4-kinase activity, particularly significant at the plasma membrane, endosomal, and Golgi compartments. Related references: PMID:11923287, PMID:12324459
- PI4K2B is involved in the production of InsP3 in stimulated cells and likely regulates vesicular trafficking, linking phosphoinositide signaling to membrane dynamics.
- The protein localizes to multiple membrane compartments, including the Golgi apparatus membrane, endoplasmic reticulum membrane, cell membrane, and early endosome membrane, as well as the cytosol.
- PI4K2B is widely expressed in human tissues, underscoring its fundamental role in cellular phosphoinositide metabolism.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant fusion protein encompassing amino acids 1–120 of human PI4K2B (NP_060793.2). This N-terminal region may be accessible for detection; consider validating epitope specificity in your experimental system.
- For Western blot (WB), a predominant band around 55 kDa is expected based on the theoretical molecular weight; post-translational modifications or isoform variants might result in additional bands.
- ELISA is a suggested application; optimize antibody dilution and coating conditions for your assay format.
- The antibody is reactive with human PI4K2B; cross-reactivity with other species or PI4K family members (e.g., PI4K2A) has not been reported. Assess specificity if required.
