  • PI3K regulatory subunits p85α (PIK3R1), p55γ (PIK3R3), and p85β (PIK3R2) are non-catalytic adapters that link the p110 catalytic subunit to activated receptor tyrosine kinases. p85α contains two SH2 domains and an SH3 domain, binding phosphorylated tyrosine residues on receptors such as FGFR, KIT, PDGFR, and integrin β2 (ITGB2), thereby mediating PI3K membrane recruitment and activation.   • The p85α subunit plays an essential role in insulin signaling, promoting glucose uptake and glycogen synthesis in insulin-sensitive tissues, and modulates the cellular response to ER stress by facilitating XBP1 isoform 2 nuclear translocation during metabolic overloading in the liver.   • These regulatory subunits exist as multiple isoforms: PIK3R1 produces isoforms p85α, p55α, and p50α via alternative splicing, while PIK3R2 encodes p85β and PIK3R3 encodes p55γ.
