Biological Background: PI3 Kinase p85 beta Function and Localization
- Encoded by the PIK3R2 gene, PI3 Kinase p85 beta (phosphatidylinositol 3-kinase regulatory subunit beta) is an 85 kDa regulatory subunit of class IA PI3K.
- It associates with the p110 catalytic subunit to form active PI3K, which phosphorylates PtdIns(4,5)P2 to produce the second messenger PIP3 at the plasma membrane.
- PIP3 recruits PH domain-containing proteins such as AKT1 and PDPK1, activating signaling cascades that drive cell growth, survival, proliferation, motility, and morphology.
- Through its SH2 domain, p85 beta binds to phosphorylated protein-tyrosine kinases (e.g., activated growth factor receptors), functioning as an adapter to localize the p110 catalytic subunit to the membrane.
- It has been shown to indirectly regulate autophagy (PubMed:23604317). Related references: PMID:23604317
- p85 beta promotes nuclear translocation of XBP1 isoform 2 under ER stress and/or insulin stimulation in the liver, thereby contributing to improved glucose tolerance.
- The protein contains SH2 and SH3 domains, is phosphorylated upon PI3K activation, and may undergo ubiquitin conjugation; mutations are associated with disease variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–85) of human p85 beta; this region may be less conserved, potentially providing isoform specificity, but cross-reactivity with p85 alpha should be considered and validated in the relevant sample system.
- For WB, use whole-cell lysates from human, mouse, or rat cell lines or tissues; the expected band is approximately 82 kDa. A positive control (e.g., HeLa or NIH/3T3 lysate) is recommended.
- IHC-P and IF/ICC may require optimization of antigen retrieval (heat-mediated citrate buffer, pH 6.0) and permeabilization (0.1% Triton X-100). Validate signal specificity using blocking peptide or knockout controls.
- In ELISA, this antibody can be used as a capture or detection reagent; pairing with a suitable p85 beta detection antibody is necessary for quantitative assays.
- Since p85 beta is a ubiquitously expressed intracellular protein, ensure proper fixation and permeabilization for IF/ICC, and consider co-staining with compartment-specific markers to confirm localization.
