Biological Background: PI3 Kinase p85 beta Function and Localization
- Encodes the phosphatidylinositol 3-kinase regulatory subunit beta (p85-beta), a key regulatory subunit of class IA PI3K.
- Forms a heterodimer with catalytic p110 subunits, phosphorylating PtdIns(4,5)P2 to generate the second messenger PIP3.
- PIP3 recruits pleckstrin homology (PH) domain-containing proteins such as AKT1 and PDPK1 to the plasma membrane, activating pathways regulating cell growth, survival, proliferation, and motility.
- Binds to activated receptor tyrosine kinases via its SH2 domain, functioning as an adapter to localize the catalytic subunit to the membrane.
- Indirectly regulates autophagy, as shown in functional studies. Related references: PMID:23604317
- Promotes nuclear translocation of XBP1 isoform 2 under ER stress or insulin stimulation in the liver, contributing to improved glucose tolerance (By similarity).
- Contains SH2 and SH3 domains and is subject to phosphorylation and ubiquitination; variants are associated with MPPH syndrome (PIK3R2-related).
- Theoretical molecular weight approximately 81.5 kDa (UniProt), migrating at ~82 kDa in SDS-PAGE.
