Biological Background: PI3 Kinase p85 beta Function and Localization
- Encoded by the PIK3R2 gene, the phosphatidylinositol 3-kinase regulatory subunit beta (also known as p85β, p85-BETA) is a key regulatory subunit of class IA PI3Ks, forming heterodimers with catalytic p110 subunits to regulate downstream signaling.
- As a regulatory subunit, it mediates the PI3K pathway by binding to phosphorylated tyrosine kinases via its SH2 domains, recruiting the catalytic subunit to the plasma membrane where it phosphorylates PtdIns(4,5)P2 to generate PIP3, a critical second messenger that activates AKT and PDK1, driving cell growth, survival, and proliferation.
- Indirectly regulates autophagy, a cellular degradation process, as demonstrated by functional studies (PubMed:23604317). Related references: PMID:23604317
- Promotes nuclear translocation of the spliced XBP1 isoform in response to ER stress and insulin signaling in the liver, contributing to improved glucose tolerance (By similarity). This highlights its role in metabolic regulation and stress responses.
- Mutations in PIK3R2 are associated with megalencephaly-polymicrogyria-polydactyly-hydrocephalus (MPPH) syndrome, a developmental brain disorder (indicated by the "Disease variant" keyword in UniProt).
- Post-translational modifications include ubiquitination (Ubl conjugation) and phosphorylation (Phosphoprotein), which may regulate its stability, activity, and interactions.
- Additional keywords: GTPase activation, protein transport, and stress response, underscoring its involvement in diverse cellular processes including cytoskeletal dynamics and intracellular trafficking.
- As a reference proteome member and target of proteomic identification, p85β is widely studied in signaling and cancer research.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning amino acids 100-200 of human p85β (O00459). For Western blot (WB), a band around 82 kDa is expected; consider using appropriate positive controls (e.g., human or rat cell lysates known to express PI3K signaling components) and validate antibody specificity with knockout or knockdown samples if available.
- For ELISA applications, optimization of coating antigen concentration and antibody dilution is recommended. Cross-reactivity with rat samples is reported, but performance in other species should be verified independently.
- Due to the role of p85β in PI3K/AKT signaling, this antibody may be useful in studying pathways related to cancer, metabolism, and neurodevelopmental disorders. Pair with phospho-specific antibodies or inhibitors to dissect activation states.
- Always pre-wet the PVDF membrane and use appropriate blocking and washing conditions to minimize background. Consider using a loading control like β-actin or GAPDH for normalization.
