Biological Background: PI3 Kinase p85 alpha/beta Function and Localization
- PI3-kinase regulatory subunit alpha (p85α; encoded by PIK3R1) is an adaptor protein that links activated receptor tyrosine kinases to the p110 catalytic subunit, enabling PI3K/AKT pathway activation. The antibody also recognizes the related p85β subunit (PIK3R2).
- Required for insulin-induced glucose uptake and glycogen synthesis in insulin-sensitive tissues, highlighting its role in metabolic regulation.
- Integrates signaling from numerous growth factor receptors, including FGFR1, FGFR2, FGFR3, FGFR4, KITLG/SCF, KIT, PDGFRA, and PDGFRB.
- Plays a role in integrin beta-2 (ITGB2) signaling, contributing to immune cell adhesion and activation. Related references: PMID:17626883 PMID:19805105 PMID:7518429
- Modulates cellular response to endoplasmic reticulum stress by promoting nuclear translocation of XBP1 isoform 2, enhancing glucose tolerance under metabolic overload in the liver. Related references: PMID:20348923
- Primarily located in the cytoplasm; dynamic recruitment to the plasma membrane is essential for signal transduction.
- Isoform 2 is expressed predominantly in skeletal muscle and brain, with lower levels in kidney and cardiac muscle; also present at the protein level.
- Subject to multiple post-translational modifications, including acetylation, phosphorylation, and ubiquitination; implicated in diseases such as dwarfism.
Experimental Guidance and Technical Tips
- The antibody is generated against a synthetic peptide likely recognizing a conserved epitope between p85α and p85β; verify specificity using knockout or knockdown controls.
- For Western blotting, bands corresponding to p85α (~85 kDa) and p85β (~81–84 kDa) may be observed; use appropriate positive lysates (e.g., HeLa, MCF-7, mouse brain) and titrate antibody concentration.
- In immunofluorescence and immunohistochemistry, optimize fixation (e.g., 4% PFA) and antigen retrieval (e.g., citrate buffer pH 6.0) based on sample type; test multiple dilutions.
- The antibody can serve as a capture or detection reagent in ELISA; perform pairing experiments with p110 or other interactors to validate performance.
- Cross-reactivity with human, mouse, and rat broadens translational study utility; always confirm detection in your specific model system.
