Biological Background: PI3 Kinase p85 alpha/beta Function and Localization
- Encoded by PIK3R1 (p85-alpha) and PIK3R2 (p85-beta), these regulatory subunits of phosphatidylinositol 3-kinase function as adaptor proteins linking the p110 catalytic subunit to activated receptor tyrosine kinases via SH2 domains.
- Binds activated protein-tyrosine kinases through its SH2 domain, mediating the membrane translocation of the p110 catalytic unit. Essential for insulin-stimulated glucose uptake and glycogen synthesis in insulin-sensitive tissues, and participates in signaling pathways downstream of FGFR, KIT, and PDGFR. Modulates ER stress responses by promoting XBP1 nuclear translocation in the liver, contributing to improved glucose tolerance. - Alternative splicing generates multiple isoforms; isoform 2 is predominantly expressed in skeletal muscle and brain, with lower levels in kidney and cardiac muscle.
- Subcellular localization is primarily cytoplasmic, where it facilitates signal transduction from membrane receptors to intracellular effectors.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitination, influencing its stability and activity. Keywords such as SH2 domain, SH3 domain, host-virus interaction, and disease variant (dwarfism) highlight its multifunctional roles.
- Acts as a host factor in viral interactions and is linked to developmental disorders such as SHORT syndrome (dwarfism).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 400–500 of human p85 alpha/beta, a region conserved in both isoforms, supporting recognition of alpha and beta variants.
- For Western blot (WB), validate the antibody in lysates from human or mouse cell lines known to express PI3K p85 (e.g., HeLa, 3T3-L1 adipocytes). Expected bands: ~42–54 kDa (predominantly p85-alpha) and ~81–84 kDa (p85-beta). Additional bands from splice isoforms or degradation products may appear; optimize lysate preparation and sample loading accordingly.
- For ELISA, consider pairing with a validated capture/detection antibody after in-house testing. Check cross-reactivity with related isoforms and use appropriate blocking and washing conditions to minimize background.
- Since cross-reactivity is listed for human and mouse, target sequence conservation should be verified if extending to other species; perform sequence alignment prior to application.
